چکیده

Background The presence of monosomy 3 in the primary tumor is widely accepted as the most reliable prognostic parameter, identified in approximately 50% of patients with primary uveal melanoma [1]. Long term studies have shown a 3-year survival rate of 40% if monosomy 3 is present, whereas tumors with normal chromosome 3 status rarely give rise to metastatic disease and have a 90% 3-year survival rate [2]. Currently, there is no effective adjuvant treatment for the patients with early stage uveal melanoma at high risk for metastatic disease (monosomy 3). The development of uveal melanoma at an immune-privileged site, the eye, made it questionable if immunotherapy would be a suitable treatment modality. Nonetheless, in this open label Phase II study we investigated the immunological responses in high risk uveal melanoma patients, selected based on monosomy 3, to dendritic cell vaccination.

TLR agonist rHP-NAP as an Adjuvant of Dendritic Cell-Based Vaccine to Enhance Anti-Melanoma Response

Background: Melanoma is a common and malignant cutaneous tumor, which is responsible for a large proportion of skin cancer deaths. Dendritic cell (DC)-based vaccines have achieved positive results in the treatment of melanoma because of their ability to induce cytotoxic response to facilitate tumor elimination. Objective: To improve the efficacy of dendritic ce...

Genetics of Uveal Melanoma and Cutaneous Melanoma: Two of a Kind?

Cutaneous melanoma and uveal melanoma both derive from melanocytes but show remarkable differences in tumorigenesis, mode of metastatic spread, genetic alterations, and therapeutic response. In this review we discuss the differences and similarities along with the genetic research techniques available and the contribution to our current understanding of melanoma. The several chromosomal aberrat...

Adjuvant Therapy of Uveal Melanoma: Current Status.

The survival of patients with uveal melanoma remains poor because of the development of metastatic disease. Adjuvant therapy after treatment of the primary tumor has been tested but has not been shown to prevent the development of metastasis. Several new approaches are being developed. Cytotoxic and immunotherapeutic regimens are being more rationally applied using tumor genetic criteria to bet...

Treatment of uveal melanoma: where are we now?

Uveal melanoma, a rare subset of melanoma, is the most common primary intraocular malignancy in adults. Despite effective primary therapy, nearly 50% of patients will develop metastatic disease. Outcomes for those with metastatic disease remain dismal due to a lack of effective therapies. The unique biology and immunology of uveal melanoma necessitates the development of dedicated management an...
